About Chronic Lymphocytic Leukemia (CLL)
Chronic lymphocytic leukemia (CLL) is a cancer of the blood and bone marrow that usually gets worse slowly. CLL is one of the most common types of leukemia in adults. It often occurs during or after middle age; it rarely occurs in children.Targeted therapy is a type of treatment that uses drugs or other substances to identify and attack specific cancer cells. Tyrosine kinase inhibitor (TKI)
This treatment blocks the enzyme, tyrosine kinase, that causes stem cells to develop into more white blood cells than the body needs.
1、 Ibrutinib
A drug used alone or with other drugs to treat adults with chronic lymphocytic leukemia, small lymphocytic lymphoma, Waldenstrom macroglobulinemia (a type of non-Hodgkin lymphoma), mantle cell lymphoma, or marginal zone lymphoma. It is also used to treat adults and children aged 1 year and older with chronic graft versus host disease. It is also being studied in the treatment of other types of cancer. Ibrutinib blocks a protein called BTK, which may help keep cancer cells from growing. It may also lower the body’s immune response.

3、Acalabrutinib
Acalabrutinib is indicated for the treatment of adults with mantle cell lymphoma (MCL) who have received at least one prior therapy, and for the treatment of adults with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL).

4、Idelalisib
Idelalisib is a second-line drug for patients whose chronic lymphocytic leukemia (CLL) has relapsed. Used in combination with rituximab, idelalisib is to be used in patients for whom rituximab alone would be considered appropriate therapy due to other existing medical conditions. It appears to be effective and leads to improvement of lymphadenopathy and splenomegaly.

5、Duvelisib
Duvelisib is indicated to treat adults with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL) who have received at least two prior therapies that did not work or stopped working. CLL is a type of cancer that begins in the white blood cells, and SLL is a type of cancer that begins mostly in the lymph nodes.

BCL2 inhibitor
This treatment blocks a protein called BCL2 which is found on some leukemia cells. This may kill leukemia cells and make them more sensitive to other anticancer drugs.
6、Venetoclax
venetoclax is indicated for adults with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL). Indication does not depend on mutation status (e. g. 17p deletion, IGHV mutation, 12+).

Overexpression of BCL-2 has been demonstrated in CLL cells where it mediates tumor cell survival and has been associated with resistance to chemotherapeutics.
